The recombinant canine CX3CL1 is a disulfide-linked homodimer. The reduced monomer comprises 362 amino acids and has a predicted molecular mass of 38.2 kDa. The apparent molecular mass of the protein is approximately 64-69 kDa in SDS-PAGE under reducing conditions.

CX3CL1 / Fractalkine / NTN蛋白背景綜述

Chemokine (C-X3-C motif) ligand 1 (CX3CL1) is a large cytokine protein containing multiple domains and is the only known member of the CX3C chemokine family. It is also commonly known as fractalkine (in humans) and neurotactin (in mice). The polypeptide structure of CXC3L1/ NTN / Fractalkine differs from the typical structure of other chemokines. Mature human CX3CL1/ NTN / Fractalkine consists of a 76 aa residue chemokine domain, a 241 aa residue stalk region containing 17 degenerate mucin-like repeats, a 19 aa residue transmembrane segment and a 37 aa residue cytoplasmic domain. The extracellular domain of human CX3CL1/ NTN / Fractalkine can be released, possibly by proteolysis at the dibasic cleavage site proximal to the membrane, to generate soluble CX3CL1/ NTN / Fractalkine. Soluble CX3CL1/ NTN / Fractalkine potently chemoattracts T cells and monocytes, while the cell-bound CX3CL1/ NTN / Fractalkine promotes strong adhesion of leukocytes to activated endothelial cells where it is primarily expressed. CX3CL1/ NTN / Fractalkine elicits its adhesive and migratory functions by interacting with the chemokine receptor CX3CR1/ NTN / Fractalkine.
