The recombinant human SIGIRR/Fc is a disulfide-linked homodimer. The reduced monomer comprises 359 amino acids and has a predicted molecular mass of 39.5 kDa. The apparent molecular mass of the protein is approximately 47-54 and 33 kDa in SDS-PAGE under reducing conditions.

SIGIRR / TIR8蛋白背景綜述
SIGIRR (single immunoglobulin IL-1 receptor-related molecule) is a member of the interleukin 1 receptor (IL-1R) superfamily, and was identified in mouse and human. Although SIGIRR shows the typical conserved motifs that characterize the IL-1R and Toll superfamily, it is structurally and functionally distinct from both. SIGIRR has only one Ig domain in its extracellular portion whereas the IL-1R family contains three Ig folds. An unusually long cytoplasmic domain is reminiscent of the structure of drosophila Toll, yet the SIGIRR peptide sequence is more closely related to IL-1RI. SIGIRR is a negative regulator for Toll-IL-1R signaling. It plays a critical role in gut homeostasis, intestinal inflammation, and colitis-associated tumorigenesis by maintaining the microbial tolerance of the colonic epithelium. SIGIRR-deficient colonic epithelial cells displayed commensal bacteria-dependent homeostatic defects, indicating that SIGIRR is critical in controlling the homeostasis and innate immune responses of the colon to enteric microflora. Moreover, SIGIRR-deficient mice developed stronger Th2 immune response and this discovery suggested that SIGIRR is important in the regulation of Th2, possibly through its impact on IL-33-ST2-mediated signaling.
